I'm using tftp-hpa, and the "root" is specified when the server is run with 
the "-s" option (as you have in one of your examples).
With this, if my dhcpd.conf has: filename "/eb-eepro100.zpxe"
(note the leading /, so it appears to be an absolute path), then the tftp 
server delivers the file /tftpboot/eb-eepro100.zpxe
