So I want a virtual machine with address 192.168.122.100 to talk to a machine with address 192.168.100.100. I have tried using route, but I ended up going round in circles as to where to put it - what the gateway should be and I'm sure it must be simpler than I'm making it.

For KVM, you will probably find it easier to use a bridged interface,
that way you can put your VMs on the same network regardless of which
machine they're on and you won't have to faff with routing.
