Prepare a fortune file, with the quote you want to add using strfile(6).
fortune(1) will then pick a random one for you. All you then have to do
is get this into your signature. I personally use cron job, but if you
really want it to change with each message, you could resort to more
exotic means, like making your .sig a FIFO, and have a daemon that
contantly runs fortune, redirected to that.

PS. Apparently kmail has a feature that lets you pick a random sig for
    each message anyway, but since I don't use kmail, I have no idea
    how you set it up.
